do you have your Arduino set up properly?
Correct board selected?
Correct processor selected?
Correct COM port selected?

everything was working the other day.
make sure no other apps (repetier host or whatever you use to control the printer) connected to printer. Disconnect or close other apps before attempt upload.

That does not mean something didn't somehow get changed - check it all to verify.
also, main power "should" be shut down, for flashing you should only need USB power to the board
and no other programs connected to the printer (close RH, or whatever you use)
